The dynamic properties of synaptic strength that support information processing in the brain depend not only on neuronal activity but also on interactions with astrocytic networks. Using the mouse hippocampus as a model, we have been investigating the molecular and cellular mechanisms that shape synaptic activity in local circuits. Our recent work focusing on the roles of astrocytes revealed two key modes of regulation: NMDA receptor–mediated signaling within astrocytes and synaptic modulation by astrocyte-derived cell adhesion molecules, including the amyloid precursor protein (APP). In this presentation, our latest findings on the physiological functions of astrocytic APP and their implications for synaptic circuit design will be discussed.
